How can I add a first class area to my plane?
I have already created a cabin profile to my plane and set it as default but it doesn't seem to be activated. The cabin configuration is always following the standard configuration.

Setting to default means that all new planes from now will get the new config automatically. Your existing planes have to be rebuild manually. Go to your fleet and select the plane where the new cabin has to be put in. At the bottom of the page you can now select the new cabin. Take notice of the costs of the cabin-rebuilding.
Will the change of cabin configuration be immediate or it has to wait for a specific amount of time?

already booked or in-flight? my presumption was that all flights after the "in-flight flight" will be affected
All flights that are already available for booking (usually the next 72h) can't be changed any more and will therefore still use the previous configuration. All new or additional flights that are entered into the system will use the new configuration.
if flights are avail to book with old cabin config and you don't really want to offer the flight with old cabon config as it might affect image and rating,
simply cancel all flights, delete the flight number from the menu: Commercial -> Flight Numbers -> Delete unused flight numbers
you can reuse the flight number to re-create the flight plan and activate it with new cabin config
