Make sure you have all the check-boxes under “apply settings to” set correctly, so the changes you make can have an actual effect.
also keep in mind that only flights not already in the system are being effected. the flights, that were already created in the system (72 hours in advance) are set and can not be changed.

Does this mean, as a main rule; that new prices apply only to flights that are planned 72 hours after the price change? Did I get it right?

Any changes will only apply for NEW flights and not for already booked flights. Hence yes, 72 hours after the change you will also see the change.
You make a price change on a route, the change is effective immediately for any new booked and scheduled flights. Already booked flights at the time of change will see change effected in 72 hours.
