I have never had a cargo company, so I can’t give you many hints.
But I know cargo companies can work worldwide, so it does not matter where you start your airline.
Every airport page shows how much traffic that airport attracts, both for passengers and for cargo. So I would fly between airports that have enough green bars for cargo. Oh, and make your hub in an airport with enough slots, or you will get stuck when you grow.

so its may be better also to start in the US, europe, china or japan
If you are new to the game, dont start with cargo. It is a hard business. I ran a big Cargo Airline myself in the past on Nicosia. You need a good concept and run many feeders to earn good money.
I suggest you start with a pax airline first. try out some planes and concepts. Have a look at the flight plans of other airlines to get an idea how the game works.
However, I mainly used the TU-204 and ATR72 in my fleet and was based in SDF and later built up a europran hub in Liege. But that was before the last big patch. Now the TU has a very long production time and is more expensive in leasing. Others used old 727C and 737C (not the new 737-700C) which worked fine too.
Why would you want others to tell you how to set up everything? There’s no such a thing as the best aircraft nor the best place to start, it all depends on YOUR mission, what YOU want to achieve and how you want to achieve it. Making good decisions is key to success. It’s not always easy, but it sure is fun and makes you feel good later if you figure out that you made a good decision, or motivates you to do better if you didn’t. Enjoy it yourself, don’t let others spoil the fun!
I had built up a hub in Sharjah just because it’s near my holding’s hub in Saudi-Arabia and Dubai was runover this time so Sharjah was likely empty because less passenger demand. Still most of my fleet is the Boeing 737-700C and Tupolev Tu-214C - more 737 just because they’re lighter and I’m more flexible and can easy rise capacities without getting in trouble, also the Tupolev has higher margins on same routes. Most traffic I have is going through my hub but not to my hub so I could have chosen one smaller airport but the start would have been much more difficult. Just 17% are going directly in my hub or from my hub. So, you need of course your hub with own significant demand for cargo (4 bars +) but this hub should also be on a good position to make transfer suitable.
Just some example:
I fly Sharjah - Doha twice daily with Boeing 737-700 (cap 2x 17 tons (34 tons)) - this airport has 4 cargo bars.
I fly Sharjah - Sialkot seven times daily with Boeing 737-700 (cap 7x 17 tons (119 tons)) - this airport has just 2 cargo bars. To say is Sialkot is a new airport and not so crowded with cargo airlines.
It’s no matter of how big but how much traffic you can get THROUGH your cargo hub, it’s even more important than with a passenger airline. You live from those CU going from B via A to C.

just 200 or up to 300 CU flying just this route
The plane choice is easy done. Go with the performance tool. Anything what needs more fuel (liters) than it can transport as cargo (kg) can written off the list. The best planes needin’ 50% fuel/kg on ~2,000km routes which is my evaluation circle and it seems to work fine for me with narrowbodies of course, widebodies ~7,000km). (e.g. 3,000 liter / 6,000kg)
But like everything in AirlineSim. You can plan like you want. Invest money like you want. At the end you have to be lucky to find the perfect place and the perfect routes for this place. Same hub with North - South-connections can be the hell, but with West - East-connections it can be heaven. Just test it. I needed about 4 years to get a well-situated cargo airline running.
I quite agree with that. Unfortunately the choice of modern cargo aircraft (ie ones you can lease) is extremely limited. The ATR42 gives you a little more flexibility, even tho the capacity is not so good the range profile is better than the 72. I’d suggest getting an IL with a carrier who does not have a monopoly in your area. For example if you are operating in Europe, look for a Far East carrier flying to Europe with big capacity aircraft.
Also, use ORS to identify routes where nobody is operating any direct cargo flights. With luck you can get a 100% load factor straight away even on airports with just a few cargo bars, and you can dictate your prices.
Cargo operations are not easy, and it’s hard to get a good profit.

If I don’t fill up an aircraft with seats, is the remaining room put to cargo automatically?
only if you have a convertible aircraft , if its not convertible you transport - nothing
And just to clarify (beacuse I have not seen it written on the Wiki), if I have towns a and b which both don’t allow transfers, and I run an aircraft from AtoB and then from B to C, passengers and freight from A that want to get to C will stay on the aircraft effectivly transfering flights, correct?
And if this is correct, what is the effective numeber of stops you can have (A-B, B+C, C+D, D+E) where passengers and or cargo from A will sitll get on the aircraft to the end of the line?
